Cross-reaction with human immunoglobulins and fetal calf serum has been removed by solid-phase absorption.
Intended use For in vitro diagnostic use. F 0479 is intended for use in flow cytometry. F 0479 has been produced to give optimal performance in flow cytometry when used in combination with a monoclonal mouse antibody. Being the F(ab')2 fragment of goat immunoglobulin, the reagent is particularly well-suited for work on unfixed cells exhibiting Fc receptors, and also for other applications where the Fc part of the antibody molecule could disturb. F 0479 is also well-suited for the screening of mouse hybridoma cell cultures for antibody production. Reagent provided F 0479 is the purified and F(ab’)2 fragmented immunoglobulin fraction of goat antiserum conjugated with fluorescein isothiocyanate isomer 1 (FITC). The conjugate is provided in liquid form in 0.05 mol/L Tris/HCl, 15 mmol/L NaN3, 1% bovine serum albumin, pH 7.2. Protein concentration g/L: See label on vial. F/P ratio: E495 nm/E278 nm = 0.65 ± 0.05 corresponding to a molar FITC/protein ratio of 2.5. Preparation 1. The immunoglobulin fraction has been solid-phase absorbed to remove antibodies cross-reacting with human immunoglobulins and fetal calf serum. 2. The absorbed immunoglobulin fraction has been further purified by affinity chromatography using agarose beads coupled with mouse immunoglobulins. 3. The affinity-isolated immunoglobulin fraction has then been degraded by pepsin and the F(ab')2 fraction isolated by gel filtration. 4. Finally, the F(ab')2 fraction is conjugated with fluorescein isothiocyanate isomer 1. Immunogen Immunoglobulins, mainly IgG, isolated from mouse serum. Specificity The antibody reacts with all mouse IgG subclasses and mouse IgM. Reaction with other mouse immunoglobulins has not been investigated, but the antibody will probably react with these via their light chains. Cross-reaction with human immunoglobulins and fetal calf serum is very low as determined by ELISA, less than 0.5%. The cross-reaction with immunoglobulins of rabbit and swine is less than 0.5%. The cross-reaction with immunoglobulins of guinea pig is about 10%, and with rat immunoglobulins about 30%. Precautions 1. For professional users. 2. This product contains sodium azide (NaN3) a chemical highly toxic in pure form. At product concentrations, though not classified as hazardous, sodium azide may react with lead and copper plumbing to form highly explosive build-ups of metal azides. Upon disposal, flush with large volumes of water to prevent metal azide build-up in plumbing. 3. As with any product derived from biological sources, proper handling procedures should be used. Storage Store in the dark at 2-8 C. Do not use after expiration date stamped on vial. If reagents are stored under any conditions other than those specified, the conditions must be verified by the user. Conjugates should not be stored in diluted form. If unexpected staining is observed which cannot be explained by variations in laboratory procedures and a problem with the antibody is suspected, contact our Technical Services. Dilution guidelines In flow cytometry, F 0479 may be used at a volume of 10 L per test and in a dilution of 1:10-1:20. This is a guideline only. The optimal conditions may vary depending on specimen and preparation method, and should be determined by each individual laboratory. Note that fluorochrome conjugates are light sensitive, and samples should be protected from light during the staining procedure and until the analysis.
