Basic ELISpot for enumeration of cells secreting IgM
Human IgM ELISpotBASIC enables enumeration of B cells secreting total human IgM as well as antigen-specific human IgM. It contains matched quantities of monoclonal capture and detection antibodies, Streptavidin-HRP and polyclonal activators.
Antigen-specific B cells can be enumerated in two alternative ways; by coating with antigen or by using biotinylated antigen in the detection step. For memory B cells, we recommend pre-stimulation with R848 and recombinant human IL-2 (both included).
Human IgM ELISpotBASIC is also available with Streptavidin-ALP. See Related products.
As complementary products, we offer ELISpot plates and substrates. See Related products.
Analyte Information
IgM
Monomeric IgM (Immunoglobulin M) is found on the surface of B cells whereas the IgM molecules secreted from plasma B cells are pentamers, i.e. five IgM molecules joined together by the J chain. IgM constitutes approximately 10% of the total Ig concentration in serum. IgM is the first antibody isotype induced in a typical immune response and hence is the isotype with the lowest affinity for antigens. Due to its structure, with 10 antigen-binding sites per IgM pentamer, it can nevertheless bind e.g. surface-bound antigens with a very high avidity.
