ELISpotPLUS for enumeration of cells secreting IFN-γ in response to SARS-CoV-2 peptides
This kit is ideal for users who want a convenient and sensitive assay. The assay is designed for the enumeration of cells secreting human IFN-γ in response to SARS-CoV-2 peptides.
The kit includes one ELISpot strip plate pre-coated with monoclonal antibody, SARS-CoV-2 S1 scanning pool, SARS-CoV-2 S2 N defined peptide pool, SARS-CoV-2 S N M O defined peptide pool, biotinylated detection antibody, Streptavidin-ALP and ready-to-use BCIP/NBT-plus substrate. A polyclonal activator for human T cells (mAb CD3-2) is supplied as a positive control for cytokine secretion. Pre-coated plates reduce assay time and minimize variability. In addition, strip-plates are ideal when running a limited number of wells.
The SARS-CoV-2 S1 scanning pool contains 166 peptides from the human SARS-CoV-2 virus. The peptides are 15-mers overlapping with 11 amino acids, covering the S1 domain of the spike S1 protein (amino acid 13-685). The pool is supplied in two vials: pool 1 SARS-CoV-2 S1 peptides 1-83 (83 peptides) and pool 2 SARS-CoV-2 S1 peptides 84-166 (83 peptides).
The SARS-CoV-2 S N M O defined peptide pool contains 47 synthetic peptides from the human SARS-CoV-2 virus. The peptides are derived from the spike, nucleoprotein, membrane protein, ORF3a and ORF7a.
The SARS-CoV-2 S2 N defined peptide pool contains 41 synthetic peptides from the human SARS-CoV-2 virus. The peptides are derived from the spike and nucleoprotein.

IFN-γ
Interferon-γ (IFN-γ, also known as IFN-gamma, IFN-g, IFNg and Interferon-gamma) is produced mainly by activated T cells and NK cells. It is a proinflammatory cytokine that activates macrophages and endothelial cells, but it also regulates immune responses by effecting APC and T and B cells. Production of IFN-γ by helper T cells as well as cytotoxic T cells is a hallmark of the TH1-type phenotype, thus, high-level production of IFN-γ is typically associated with effective host defense against intracellular pathogens.
