Poseidon™ AURKA (20q13) & 20q11 probe

Introduction: The mitotic kinase-encoding gene AURKA (STK15; STK6) is associated with aneuploidy and transformation when overexpressed in mammalian cells. The presence of AURKA gene overepression and amplification has been shown in breast, ovarian, and bladder cancer.

Intended use: The AURKA (20q13) specific DNA Probe is optimized to detect copy numbers of the AURKA gene region at region 20q13. The 20q11 specific DNA probe is included to facilitate chromosome identification. The probe is recommended to be used in combination with one of the Poseidon Pretreatment kits providing necessary reagents to perform FISH on various sample types for optimal results. Critical region 1 (red): The AURKA (20q13) gene region probe is direct-labeled with PlatinumBright550.

Control region 2 (green): The 20q11 control DNA probe is direct-labeled with PlatinumBright495.

Reagent: Poseidon probes are direct-labeled DNA probes provided in a ready-to-use format. Apply 10 μl of probe to a sample area of approximately 22 x 22 mm. Please refer to the Instructions for Use for the entire Poseidon FISH protocol.

Interpretation: The AURKA (20q13) probe is designed as a dual-color assay to detect amplification at 20q13. Amplification involving the AURKA gene region at 20q13 will show several red signals, while the control at the chromosome 20q11 specific region will provide 2 signals. Two single color red (R) and green (G) signals will identify the normal chromosomes 20 (2R2G).
